Ipsen Secures First ADC with $900M Deal for ROR1-Targeting STRO-003

April 2, 2024, IpsenIpsen Announces Collaboration with Sutro Biopharma on Solid Tumor CandidatesROR1 ADC（STRO-003) reached a cooperation.

The former obtains global exclusive rights for the development and commercialization of STRO-003, while the latter is eligible to receive a near-term payment of $90 million (including equity financing) as well as a total amount ofUp to$900 million in potential milestone payments.

ROR1It is considered a new drug target with broad-spectrum anti-cancer potential, and is mostly lowly expressed or not expressed in normal cells.Overexpressed in hematological malignancies such as B-cell lymphoma, inWidely expressed in solid tumors such as triple-negative breast cancer and ovarian cancer.

According to preclinical data, STRO-003 demonstrated anti-tumor activity in non-small cell lung cancer (NSCLC) and breast cancer models; the drug also showed good tolerability in animal models.Compared to other TOPO-1 inhibitors(Topoisomerase I)ADC, withPossibility of potentially reducing pulmonary toxicity.

Thus, Ipsen has obtained its first ADC drug. According to the agreement, Ipsen will be responsible for Phase I preparatory activities, including the submission of the IND, as well as all subsequent clinical development activities and global commercialization activities.Sutro has also taken this opportunity to outsource almost all products in the pipeline.

As of the cutoff date, across ChinaThe ROR1 ADC with the fastest clinical progress is Merck. In November 2020, Merck acquired VelosBio for $2.75 billion, obtaining its core pipeline VLS-101.

VLS-101 targets ROR1Composed of a humanized monoclonal antibody (a proteolytic linker) and monomethyl auristatin E (MMAE) (an anti-microtubule cytotoxin).

Currently, the drug is undergoing multiple Phase II/III clinical trials globally. Among them, a Phase II/III multicenter clinical trial targeting relapsed or refractory diffuse large B-cell lymphoma (DLBCL) was initiated in December 2021.

In addition, there are BI, Cornerstone, and KeSeveral pharmaceutical companies, including Lunbotech, Exelixis, and Bright Biology, have laid out plans for ROR1 ADC. Notably, CStone has introduced from LegoChem.CS5001(Original Research CodeLCB71), the first ADC from Cornerstone and the first ROR1 ADC in China.

202In October 0, one month earlier than Merck, CStone Pharmaceuticals introduced the global rights (excluding Korea) to LCB71 for a $10 million upfront payment and $353 million in milestone payments. The drug is currently in Phase 1 clinical trials and completed its first dosing in China in April 2023.

Southwest Securities research report predicts that the market potential for ROR1 ADC could reach tens of billions of dollars. Moreover, there are limited players in this field.
