Type 2 Diabetes and Obesity: Metabolic Diseases Threatening Human HealthType 2 diabetes is often accompanied by obesity. Existing incretin analogs, such as glucagon-like peptide-1 (GLP-1), exhibit significant blood sugar-lowering effects but have suboptimal weight loss outcomes and are associated with side effects like nausea and vomiting. GLP-1/glucose-dependent insulinotropic polypeptide (GIP) dual-agonists demonstrate synergistic effects in regulating blood glucose and reducing body weight, while also improving lipid homeostasis and minimizing adverse reactions such as nausea and vomiting, showing great promise in the treatment of type 2 diabetes and obesity.
GLP-1R/GIPR Dual Agonist DR10627 is a chemically synthesized small peptide containing 39 amino acids. It introduces GIP activity-related mutation sites based on the GLP-1 sequence and cross-links long-chain fatty acids to extend the drug's half-life. Preclinical research results indicate that DR10627 exhibits good GLP-1R and GIPR agonistic activity in vitro, demonstrates favorable pharmacokinetic characteristics in cynomolgus monkeys, and effectively reduces blood glucose levels in db/db mice while improving body weight and lipid metabolism in DIO rats.
