Daiichi Sankyo and AstraZeneca Announce Positive Topline Results from DESTINY-Breast06 Phase III Trial of Enhertu in HR-Positive, HER2-Low Metastatic Breast Cancer

The study results showed that, in the main trial population, patients with HR-positive, HER2-low (IHC 1+ or IHC 2+/ISH-) metastatic breast cancer who had received one or more endocrine therapies demonstrated a statistically significant and clinically meaningful improvement in PFS with ENHERTU compared to standard-of-care chemotherapy. Additionally, an improvement in PFS was also observed in the overall population, i.e., in patients with HER2-low and HER2-ultralow (IHC 0; IHC >0 <1+) metastatic breast cancer.

Although OS data are not yet mature, an early trend toward OS improvement has been observed in both HER2-low breast cancer patients and the overall population compared to standard-of-care chemotherapy. The trial will continue to further evaluate OS and other secondary endpoints. Safety findings were consistent with those observed in previous trials, with no new safety signals identified. Detailed results will be presented at an upcoming medical conference.

DESTINY-Breast06(DB06) is a Phase III clinical trial for HR-positive, HER2-low breast cancer, enrolling patients with metastatic breast cancer who have progressed after endocrine therapy and have not received chemotherapy (post ET, chemo-naive).

The DB04 trial, which previously achieved positive results, targeted patients regardless of HR status who had undergone prior endocrine therapy and one to two lines of chemotherapy.Positive results achieved on DB06 indicate a shift towards earlier treatment lines in HER2-low breast cancer.
