▎WuXi AppTec Content Team Report
On May 8, SANEGENEBIO announced its co-development with Innovent BioSGB-3908 Injection, a Small Nucleic Acid (siRNA) Drug for the Treatment of HypertensionThe clinical trial application has been officially accepted by the Center for Drug Evaluation (CDE) of the China National Medical Products Administration (NMPA) in recent days.
Hypertension is a common chronic disease that not only increases the risk of cardiovascular and cerebrovascular diseases but also may lead to complications such as kidney damage and vision loss. With the aging population and the prevalence of risk factors like obesity, lack of physical activity, and unhealthy diet, the global prevalence of hypertension continues to rise. Although there are currently effective clinical treatments for lowering blood pressure, antihypertensive medications need to be taken daily. Since hypertension is not easily detectable when symptoms are mild, patients may forget to take their medication, and poor adherence has become a major challenge in the treatment of hypertension.BloodAngiotensinogen (AGT)It is the most upstream precursor of the renin-angiotensin-aldosterone system (RAAS). The role of this target in blood pressure (BP) regulation has been confirmed, and inhibiting AGT has recognized antihypertensive effects.. For the treatment bottleneck of hypertension, siRNA drugs can directly reduce the expression of AGT, offering differentiated advantages such as long-lasting drug effects, good safety, and high patient compliance, which are expected to bring better treatment options and long-term benefits to patients.SGB-3908 is an siRNA drug targeting AGT, intended for the treatment of hypertension.According toSANEGENEBIO Press Release,SGB-3908 CollectionOf the companyA new generation of siRNA drug platform technology enables drugs to have superior activity and prolonged efficacy, with good safety and tolerability.Preclinical experimental data show that SGB-3908 can reduce hypertensionThe levels of AGT protein and related biomarkers (ANG I, ANG II) in cynomolgus monkey serum significantly decreased, achieving a pronounced blood pressure-lowering effect. The effect was long-lasting, with no safety issues such as hypotension observed. In December 2023, SANEGENEBIO and Innovent Bio announced a strategic collaboration agreement to jointly develop the siRNA candidate drug SGB-3908 for the treatment of hypertension. Meanwhile, Innovent Bio has obtained an exclusive option for the future development, manufacturing, and commercialization of this drug.Dr. Wang Weimin, Founder and CEO of SANEGENEBIO, stated: "We are very pleased that SGB-3908 has reached another significant milestone, demonstrating the execution efficiency of the SANEGENEBIO team and further enriching the company’s innovative drug pipeline in the cardiovascular and metabolic disease areas. The rapid progress of this project is due to the close collaboration and strong cooperation between both teams over the past few months. We look forward to maintaining this excellent spirit of cooperation, accelerating the clinical trials of SGB-3908, achieving positive progress, and benefiting a large number of hypertension patients as soon as possible with more and better treatment options."
