GCC19CART is an autologous CAR-T candidate product specifically designed to target GCC for the treatment of advanced colorectal cancer.A multi-center Phase I dose-escalation clinical trial initiated by researchers in China showed that, according to the Response Evaluation Criteria In Solid Tumors (RECIST 1.1), the objective response rate (ORR) was 15.4% (2/13) in the low-dose group (1x10^6 CAR-T/kg) and 50% (4/8) in the medium-dose group (2x10^6 CAR-T/kg) for GCC19CART.。
In August 2022, GCC19CART initiated a Phase I clinical trial in the United States. This clinical trialConducted in collaboration with several renowned medical centers in the United States, includingDana FArber (Dana-Farber Cancer Institute, Harvard University),UCSF (University of California, San Francisco) Cancer Center,University of Colorado (Cancer Center) and COH (City of Hope Cancer Center).As of November 1, 2023, low-doseThe dosage group (1x10^6 CAR-T/kg) enrolled four patients, who have received reinfusion treatment and completed the evaluation.According to RECIST1.1 criteria, as independently evaluated by a U.S.-based third party, two patients in the low-dose group achieved partial response (PR), with an ORR of 50%.(The resultBetter than the clinical research data in China at the same dose level), the clinical efficacy is much superior to the third-line standard therapy approved by the U.S. FDA (ORR is only 1%-1.6%).. The clinical trial of the low-dose group has been completed,The clinical trial of the medium-dose group (2x10^6 CAR T/kg) is currently underway. As of April 18, 2024, three patients in the medium-dose group have completed the infusion treatment, two patients have completed the evaluation, and one patient has achieved PR.
Preliminary results from the U.S. Phase I clinical trial indicate that GCC19CART demonstrates significant clinical efficacy and favorable safety in recurrent/refractory metastatic colorectal cancer.Combined with clinical research data in China, it can be seen that clinical efficacy is positively correlated with dosage.The trial is still ongoing, Clinical Trial Registration Number:NCT05319314。
