Bristol Myers Squibb recently announced the latest 4-year results from the POETYK PSO Long-Term Extension (LTE) trial of Sotyktu (deucravacitinib), a selective tyrosine kinase 2 (TYK2) inhibitor, for the treatment of adult patients with moderate to severe plaque psoriasis. The data showsAt week 208, the response rates for PASI 75 (at least 75% improvement in Psoriasis Area and Severity Index) and PASI 90 were 71.7% and 47.5%, respectively, and the response rate for sPGA 0/1 (static Physician Global Assessment of clear or almost clear skin) was 57.2%.These data were announced at the recent EADV Spring Symposium held by the European Academy of Dermatology and Venereology (EADV).

Sotyktu is an oral, selective TYK2 allosteric inhibitor with a unique mechanism of action.Scientists at Bristol-Myers Squibb designed Sotyktu to selectively target TYK2, thereby inhibiting the signaling of IL-23, IL-12, and type I interferons (IFN), which are key cytokines involved in the pathogenesis of various immune-mediated diseases.Sotyktu achieves allosteric inhibition of TYK2 and its downstream functions by highly selective binding to the regulatory domain of TYK2. At therapeutic doses, Sotyktu does not inhibit JAK1, JAK2, or JAK3.
