On June 5, according to the CDE official website, Pfizer's Class 1 chemical drug PF-07820435 tablets received clinical trial implied permission, with the indication for advanced or metastatic solid tumors. Public data shows that this drug is a STING agonist.

STING (Stimulator of Interferon Genes) is a key signal transduction molecule involved in innate immune responses, primarily triggered by cytoplasmic DNA from both pathogens and hosts. STING agonists hold potential for treating pathogen infections and cancer, as this factor is widely expressed in various cell populations, including cancer cells, with functions such as activating Type I interferons, inducing autophagy, or promoting apoptosis. Particularly for cancer patients who do not respond well to anti-PD-1 monotherapy, STING agonists represent a promising immunotherapy approach.

PF-07820435 is a STING agonist developed by Pfizer, currently undergoing Phase I clinical trials overseas. This study aims to evaluate the early signals of safety and anti-tumor activity of PF-07820435 when administered alone (Part 1A) or in combination with sasanlimab (Part 1B; Part 2). The first part is a dose-escalation study, while the second part will further assess the combination expansion of PF-07820435 at the recommended dose in selected patients with advanced solid tumors. Tumors involved include melanoma, squamous cell carcinoma of the head and neck, etc.

The clinical trial application for PF-07820435 was accepted by the CDE in March this year. After receiving implied approval this time, Pfizer may initiate further research in China.
