▲ Figure 1 Source:Journal "Cancer Discovery"The article confirmed that Pan Cancer T B.V. developed a targetedRopporin-1(ROPN1)TCR-T Cell Therapy in the Treatment ofTriple-Negative Breast Cancer(triple negative breast cancer, TNBC) has demonstrated preclinical safety and efficacy.This is the first time ROPN1 has been applied as a therapeutic target for breast cancer.Pan Cancer T B.V. discovered,ROPN1 is hardly expressed in healthy tissues but shows abundant and uniform expression levels in primary and metastatic TNBC.In approximately 75% of primary TNBC, ROPN1 protein can be detected in 50-100% of tumor cells. This level of abundance, expression, and homogeneity is unique among other targets in adoptive cell therapy (ACT).The surprising abundance and homogeneity of ROPN1 in TNBC provide strong support for ACT treatment.High expression and homogeneity of ROPN1 may help reduce the risk of tumor recurrence after cell therapy.Moreover, recent studies have also indicated that ROPN1 may act as an oncogene, driving tumor metastasis in vivo, and the expression of ROPN1 is negatively correlated with the survival of TNBC patients.This further highlights the great therapeutic potential of ROPN1 as an ACT target.TCR-T cell therapy genetically engineers a patient's own T cells to express specific cancer-related antigen T cell receptors (TCRs), creating TCR-T cells, which are primarily used for the treatment of solid tumors. On August 1, 2024, the world's first TCR-T therapy — TECELRA® (afamitresgene autoleucel) from Adaptimmune — was approved by the U.S. Food and Drug Administration (FDA) for marketing, intended for the treatment of advanced synovial sarcoma.Breast cancer ranks among the top in terms of incidence and mortality rates in women globally. Current treatment methods mainly include surgery, chemotherapy, radiotherapy, endocrine therapy, and targeted therapy.Among them, triple-negative breast cancer has a high degree of malignancy. In the past, chemotherapy was the main treatment method, and the prognosis was poor.Currently, Pan Cancer T is developing a broad pipeline of TCR therapies targeting 10 major solid tumors. The leading clinical program, PCT-1, has demonstrated safety and efficacy in several tumor models, with indications primarily for triple-negative breast cancer and melanoma.
▲ Figure 2: Overview of Pan Cancer T PipelinePan Cancer T's TCR-T aims to address two major obstacles in the field: the first being the universality and stability of the target,Pan Cancer T studied 30 unexplored targets that are specifically and robustly expressed in various solid tumors.; Secondly, Pan Cancer T's TCR-T is meticulously designed to enhance durability, thereby driving deeper and more sustained clinical responses across multiple solid tumors.On July 11, 2024, Pan Cancer T successfully completed a seed funding round of 4.25 million euros.EyePreviously, the total financing of Pan Cancer T has reached approximately11 million euros。Pan Cancer T plans to secure Series A financing within the next year and seeks to establish partnerships with other biotech companies or pharmaceutical enterprises to further develop its TCR-T products.Through the seed financing round, Pan Cancer T will complete the preclinical research of PCT1: CO-STIM and advance CMC activities. This funding will also support the progress of the early pipeline targeting colorectal cancer and ovarian cancer, as well as the development of a second TCR engineering approach.References:
