Prostate cancer is one of the most common malignant tumors in the male urinary and reproductive systems, significantly impacting male health and quality of life. Prostate cancer is considered an 'immune desert' because its unique tumor microenvironment makes it difficult for immunotherapy to be effective. Although traditional treatments such as surgery, radiation therapy, and androgen deprivation therapy show positive results in the early stages, the treatment outcomes for advanced and metastatic prostate cancer are often unsatisfactory.
Recently, Professor Yin Xiushan's team from ROC ROCK Biotechnology, Professor Liang Chao's team from Southern University of Science and Technology, and Professor Zhang Luo's team jointly published a research paper titled "PSMA-specific CAR-engineered macrophages for therapy of prostate cancer" on the preprint website bioRixv. The article describes the construction of CAR-M cells targeting prostate cancer by introducing PSMA-specific single-chain variable fragments and co-stimulatory domains into macrophages. Their potent anti-tumor activity and safety were validated in both in vitro and in vivo experiments, establishing a solid preclinical foundation for subsequent advancement into clinical trials.

The study found that PSMA-targeted CAR-M cells demonstrated good safety and efficacy in in vivo experiments. Histopathological analysis and biochemical test results showed that CAR-M cells did not cause significant damage to normal tissues, further verifying their safety for clinical application. Moreover, the consistent anti-tumor effects observed both in vitro and in vivo suggest that CAR-M cells hold great promise for the treatment of prostate cancer.

This study is the world's first CAR-M cell therapy regimen for the treatment of prostate cancer, laying the foundation for the application of CAR-M cells in advanced pancreatic cancer treatment. PSMA-targeted CAR-M cells demonstrated significant anti-tumor effects and good safety in prostate cancer models. In the future, ROC ROCK Biotechnology will evaluate the safety and efficacy of CAR-M for prostate cancer in clinical applications.
ROC ROCK Biotechnology is a clinical-stage cell therapy company focused on the research of macrophage-based drugs for cancer treatment. In 2023, ROC ROCK Biotechnology, in collaboration with the Affiliated Hospital of Xuzhou Medical University, initiated an Investigator Initiated Trial (IIT) for the RR-M01 pipeline. Multiple subjects have completed dosing evaluations, demonstrating that the leading pipeline RR-M01 has favorable drug safety and efficacy, bringing new hope for the treatment of malignant solid tumors, particularly recurrent and refractory ovarian cancer. The RR-M01 pipeline is expected to complete enrollment and dosing of the final patient by October 2024.
At the same time, the indications for pipeline RR-M01 have been further expanded to include all solid tumors with HER2 positivity or low expression. Ethical approval was obtained from the Affiliated Hospital of Xuzhou Medical University in early March 2024, and four subjects have been dosed simultaneously. The plan for 2024 is to recruit 20 patients. Clinical trials targeting pancreatic cancer and small cell lung cancer are also proceeding smoothly.
