Senti Biosciences Reports Positive Phase 1 Data for Off-the-Shelf CAR-NK Therapy SENTI-202 in Relapsed/Refractory AML, Shares Surge 439%

Recently, Senti Biosciences, Inc. (NASDAQ: SNTI) announced preliminary data from a Phase I clinical trial targeting relapsed/refractory hematologic malignancies, including acute myeloid leukemia (AML). The therapy, named SENTI-202, is an off-the-shelf CAR-NK cell therapy designed to selectively target and eliminate hematologic malignancies expressing CD33 and/or FLT3 while sparing healthy bone marrow cells.

As of the data cutoff for this cohort, among the three AML patients treated at the lowest dose level (1 billion CAR+ NK cells per dose), two achieved complete remission (CR), with both patients testing negative for minimal residual disease (MRD), indicating no detectable cancer cells in their bone marrow samples. At the time of data analysis summary, these two patients had maintained a disease-free status for over 4 months and 3 months, respectively. All three patients generally tolerated SENTI-202 well, with an adverse event profile consistent with AML patients receiving conventional chemotherapy. Additionally, the transgene of SENTI-202 was detectable in the peripheral blood of all three patients across all treatment cycles, exhibiting pharmacokinetic characteristics consistent with other experimental CAR-NK therapies.

Currently, this Phase I clinical trial is planning to conduct dose escalation with a higher dose of 1.5 billion CAR+ NK cells/dose, with more data on safety and efficacy, including initial persistence data, expected to be reported by 2025.

Impacted by the aforementioned positive clinical data, the share price of Senti Biosciences soared on December 2, 2024, with a trading volume reaching 73.4 million shares, far exceeding the average trading volume of 17,810 shares.The stock price surged by 439%., closing price was $11.66 per share.

SENTI-202 is a universal CAR-NK cell therapy candidate product designed using the synthetic biology technology platform Gene Circuits. It aims to selectively target and eliminate hematologic malignancies expressing CD33 and/or FLT3, such as AML and myelodysplastic syndrome (MDS), while preserving healthy bone marrow cells. It consists of three main components: an activating CAR for recognizing CD33 and FLT3, an inhibitory CAR for protecting healthy cells, and a calibrated IL-15 release mechanism to enhance cell persistence and activity.
