CAR-T cell therapy has achieved remarkable success in the treatment of hematological tumors, but it still faces many challenges in solid tumors. Among them, the most prominent issue isOn-target Off-tumor ToxicitySince most of the targets for solid tumors are tumor-associated antigens (TAA), which are also expressed in normal tissues, CAR-T cells may mistakenly attack normal tissues, leading to severe toxic side effects. To address this issue, Immunofoco, a Suzhou-based biotechnology company specializing in cell therapy for solid tumors, has collaborated with Professor Bo Zhai from Renji Hospital affiliated with Shanghai Jiao Tong University (currently the Director of the Minimally Invasive Interventional Treatment Center at Dongfang Hospital affiliated with Tongji University), and other teams to develop Peri Cruiser.®Innovative technology platform, by adjusting the tissue distribution of CAR-T cells in the body,Significantly reduced the on-target off-tumor toxicity caused by the expression of tumor-associated antigens (TAA) in normal tissues. This platform not only retains the ability of CAR-T to kill primary tumors but also effectively controls tumor metastasis.Provides a breakthrough solution for the application of CAR-T in solid tumor treatment. This research achievement was published on January 22 in a top academic journal.Science Translational Medicine, and featured as the cover article of that issue, marking an important breakthrough for the company in the field of solid tumor CAR-T cell therapy!

Peri Cruiser®Technology platform based onMultiple shRNA TechnologySimultaneously achieving CAR expression and knocking down the expression of multiple key T cell adhesion and migration molecules on a lentiviral vector can significantly reduce the toxicity of CAR-T cells in normal tissues. Study results show that this technology can significantly decrease CAR-T entry into normal tissues (e.g., liver and lung) while preserving the ability to infiltrate tumors. Through various in vitro experiments, such as continuous killing assays, it was demonstrated that reducing the expression of adhesion molecules can enhance the sustained cytotoxic capacity of CAR-T cells against tumors, decrease the expression of exhaustion markers, and increase the proportion of memory cells. Meanwhile, it does not affect the formation of immunological synapses between CAR-T and tumor cells, showcasing the multiple advantages of this technical platform. Researchers also conducted in vivo efficacy studies in colon cancer and gastric cancer models, Peri Cruiser®The technical platform retains the ability to suppress various solid tumors and metastatic tumors, demonstrating overall good anti-tumor activity. This research not only presents a completely new CAR-T technology concept but also provides solutions for safer and more effective application of CAR-T therapy in solid tumors.
