According to the "Report on the Incidence and Mortality of Malignant Tumors in China" released by the National Cancer Center in December 2024, the annual number of deaths from the three major gastrointestinal tumors—esophageal cancer, stomach cancer, and colorectal cancer—reached 687,900, accounting for 26.7% of the total number of deaths from all malignant tumors. Facing the severe trend in the field of gastrointestinal tumors, issues related to prevention, diagnosis, and treatment have remained focal points within the industry. In terms of treatment, groundbreaking innovative therapeutic drugs have successively been approved for clinical use, bringing new hope to patients with gastrointestinal tumors.

Recently, the clinical trial application for DR30206 Injection, a Class 1 new biological product developed by Zhejiang Doer Biologics Co., Ltd. (hereinafter referred to as "Doer Biologics"), for use in combination with standard chemotherapy in patients with advanced or metastatic gastrointestinal tumors has been approved. This marks the second approved clinical indication for DR30206, aiming to evaluate the safety, tolerability, and preliminary efficacy of DR30206 in combination with standard chemotherapy regimens in gastrointestinal tumors.Previously, the product had also been approved for the indication of advanced solid tumors. The approval of the new indication marks another significant milestone in the development process of this innovative drug.

Gastrointestinal Tumor Drugs in Rapid Development

In recent years, immunotherapy and targeted therapy technologies have rapidly developed in the treatment of gastrointestinal tumors. PD-1/PD-L1 antibodies and some targeted therapy drugs have been approved by the U.S. Food and Drug Administration (FDA) and China's National Medical Products Administration (NMPA) for the treatment of advanced gastrointestinal tumors, which has also driven the continuous development of more related innovative drugs. However, compared with lung cancer, melanoma, etc., the research and development progress of innovative drugs for gastrointestinal tumors globally still requires ongoing effort.

Innovations Originating from China

DR30206 for Injection is a Class 1 therapeutic biologic independently developed by Doer Biologics with global intellectual property rights. It is an antibody fusion protein targeting PD-L1, VEGF, and TGF-β. This innovative tri-specific drug is designed, modified, and developed based on Doer Biologics' proprietary SMART-VHHBody and MultipleBody® platform technologies. It exhibits high affinity and excellent specificity for the three targets: PD-L1, VEGF, and TGF-β. Hitting three targets simultaneously is no small feat. To date, globally, there are no antibody fusion protein drugs targeting PD-L1, VEGF, and TGF-β that have been marketed.

In terms of mechanism of action, DR30206 restores the proliferation of exhausted CD8+ T cells by blocking the PD-1/PD-L1 signaling pathway; it also reduces tumor angiogenesis and alleviates immunosuppression by specifically binding to free VEGF and TGF-β, thereby achieving the purpose of killing tumors. The results of completed non-clinical studies and ongoing Phase I clinical trials show that DR30206 has a clear mechanism of action and anti-tumor growth effects, with good safety, supporting the initiation of clinical trials for DR30206 in combination with standard chemotherapy in patients with advanced or metastatic gastrointestinal tumors.
