Renal cell carcinoma is a malignant tumor originating from the renal tubular epithelium, accounting for 80%-90% of renal malignant tumors. The incidence rate of renal cell carcinoma in China is 4.02/100,000, and the age-standardized incidence rate is 2.66/100,000. Renal cell carcinoma is an aggressive malignant tumor with insidious onset, showing almost no symptoms in the early stages. According to statistics, approximately 30% of patients with renal cell carcinoma have already developed cancer metastasis at the time of diagnosis, missing the optimal timing for surgery; among those who can undergo radical nephrectomy, 25% will experience local recurrence or distant metastasis after the surgery, leading to poor prognosis for patients with recurrent or metastatic renal cell carcinoma. There is an urgent need to design new clinical trials and develop innovative treatment strategies.
Pazopanib is a multi-target tyrosine kinase inhibitor that primarily acts on pathways such as VEGFR, PDGFR, and c-Kit, inhibiting angiogenesis and tumor growth. In 2017, pazopanib was launched in China and approved for first-line treatment of patients with advanced renal cell carcinoma and for patients with advanced renal cell carcinoma who had previously received cytokine therapy. Its serious adverse reaction is hepatotoxicity, which has been included in the black box warning in the package insert. Cell therapy, as a "living drug," has already achieved cures in certain hematologic malignancies. The scientific basis for the synergistic effect of pazopanib and NK cells mainly comes from preliminary studies and some research on renal cell carcinoma.(RCC), soft tissue sarcoma, and other cancers. These studies have shown that pazopanib modulates the tumor microenvironment and enhances the anti-tumor effects of NK cells. IBR854 cell injection combined with pazopanib is expected to bring new hope to patients in the treatment of renal cell carcinoma.

IBR854 Cell Injection is a globally innovative product developed by Imbioray that combines 5T4 antibodies with NK cells through conjugation. It is the first ACC-NK cell therapy in China for solid tumors using non-viral vectors and non-genetic modification methods. The core structure of this product involves covalently conjugating a specific antibody targeting the tumor antigen 5T4 to allogeneic NK cells via a linker. The 5T4 antigen is a highly glycosylated cell surface protein found in human placental trophoblasts and various types of human cancer cells. It shows high positive expression rates in multiple solid tumors, including renal cell carcinoma, epithelial ovarian cancer, non-small cell lung cancer, breast cancer, pancreatic cancer, cervical cancer, soft tissue sarcoma, prostate cancer, and colorectal cancer, but it is not expressed in normal tissues of healthy adults. The broad distribution of 5T4 across various malignant tumors makes it a popular target for cancer immunotherapy.
