Grit Bio collaborates with Tianze Cloud to pioneer a new generation of mRNA tumor vaccine delivery system, which can selectively and precisely target antigen-presenting cells (APCs) throughout the body, significantly enhancing immune response while reducing toxicity. The research team has developed a novel APC-targeting lipid nanoparticle (APC-LNP) delivery system that achieves efficient and specific transfection of APCs under intravenous administration, with minimal impact on liver and lung parenchymal cells.

Preclinical studies show that the dose of APC-LNP required to induce an equally potent antigen-specific T-cell response is only one-tenth of the LPX (lipoplex, a systemic APC-targeting delivery system developed by BioNTech and already in clinical use), and APC-LNP can induce long-lasting memory responses favorable for vaccines.

Animal model data further indicates that the platform has good clinical application potential. NEOvigator™, a neoantigen discovery and prediction platform independently developed by Gracell Biotechnologies, integrates artificial intelligence and machine learning algorithms to successfully predict 20 highly immunogenic tumor neoantigens in mouse tumors. The neoantigen tumor vaccine prepared via APC-LNP/mRNA packaging achieved complete tumor suppression in a CT26 lung metastasis model and effectively prevented recurrence upon tumor re-challenge. Moreover, the neoantigen tumor vaccine also demonstrated significant efficacy in the highly malignant B16-F10 melanoma model. Preclinical safety study results obtained from multiple animal models confirmed that the system has excellent in vivo tolerance. Currently, GT601, a neoantigen tumor vaccine pipeline developed by Gracell Biotechnologies, is undergoing investigator-initiated clinical trials in China.IIT) Evaluation.
