Gravel Biotech's R&D Pipeline Layout:
1. Autologous TIL Pipeline:
(1) GT101 Injection: The first autologous Tumor Infiltrating Lymphocyte (TIL) product in China to be approved for clinical trials, currently in a pivotal clinical stage. Independently developed by the company for second-line treatment of cervical cancer, it is expected to become the first commercialized TIL therapy in China.
2. Gene-Edited Next-Generation TIL Pipeline:
(1) GT201 Injection: The world’s first membrane-bound IL-15 complex engineered TIL drug, which enhances T-cell persistence and tumor-killing ability through genetic engineering. Clinical trials have been conducted simultaneously in China and the United States, showing excellent efficacy across multiple tumor types.
(2) GT307: An iteratively innovative dual-gene knockout edited TIL therapy. GT307 demonstrates significantly enhanced anti-tumor activity by knocking out two novel key immune regulatory genes, resisting T-cell exhaustion and multiple immunosuppressive mechanisms within the tumor microenvironment, while reducing TIL dependency on IL-2 support. Leveraging its self-developed high-throughput CRISPR screening platform ImmuT Finder®, Grail Bio discovered and validated a series of key immune regulatory targets influencing the mechanism of TIL action. Preliminary clinical data for GT307 has shown good safety, with patients having "cold tumors" potentially benefiting from this drug.
3. Neoantigen tumor vaccine and off-the-shelf cell therapy product pipeline:
(1) GT601: An mRNA vaccine targeting neoantigens in tumors. This project utilizes Grail Bio's self-developed and functionally validated high-throughput sequencing combined with AI algorithms to achieve efficient prediction and identification of neoantigens, with a prediction accuracy 10 times higher than common neoantigen prediction technologies. It also employs a specially optimized APC-LNP delivery system for targeted delivery to antigen-presenting cells. Currently, GT601 is preparing to initiate IIT clinical trials.
(2) GT719: The world's first universal CAR-iNKT cell therapy derived from umbilical cord blood stem cells with clinical data. GT719 is currently in the IIT phase, exploring its efficacy in both hematological malignancies and autoimmune diseases, with preliminary safety and efficacy signals showing promising results.
(3)GT801: The Next Generationin vivo CAR-TProduct.GT801 Adopting new type T Cell-Targeting Lipid Nanoparticles (T-LNP) system, precise delivery of contents including anti-CD19 CARGeneticmRNAToTCells, subsequently produced in the bodyanti-CD19 CAR-TCell.
