We used cryopreserved PBMCs (peripheral blood-derived) as seed cells, performed activation culture in T75 culture flasks, and transferred 2E7 viable cells to a 2L perfusion culture bag after 7 days for continuous perfusion culture expansion in a scalable rocking bioreactor WAVE.Ten days later, the cell density reached 2E7/mL, with over 20 billion cells harvested. At the time of harvest, the NK cell purity was above 95%, maintaining high killing efficiency against K562.


Total Cell Expansion Performance and Cell Viability

PBMC cells were cultured with NK cell activation medium for one week and then transferred to a WAVE perfusion culture for 10 days. The cell expansion results showed that ACROBio's NK cell expansion medium could efficiently support cell growth while maintaining high cell viability.

PBMC cells were cultured with NK cell activation medium for one week and then transferred to a WAVE perfusion culture for 10 days. Flow cytometry results showed that ACROBio's NK cell culture medium could activate and maintain a high proportion of NK cells in culture, with a sustained high expression of CD16 positivity.
NK Cell Activation Markers and Inhibition Markers

PBMC cells were cultured with NK cell activation medium for one week and then transferred to a WAVE perfusion culture for 10 days. Flow cytometry results showed that ACROBio's NK cell culture medium could activate the expression of NK surface activation markers and maintain relatively low levels of inhibitory markers.
NK Cell Cytotoxicity In Vitro

PBMC cells were cultured with NK cell activation medium for one week and then transferred to WAVE perfusion culture for 10 days. In vitro killing results showed that NK cells cultured with ACROBio's NK cell medium efficiently killed K562 cells, demonstrating the ability to effectively kill target cells in vitro.
