Efficacy and Safety of FKC289, a BCMA-CD19 dual-targeted CAR-T therapy for refractory or relapsed PLA2R-associated Membranous Nephropathy

Research Background:Membranous nephropathy (MN) is one of the common causes of nephrotic syndrome in adults. Approximately 30–40% of patients with MN do not respond to initial immunosuppressive therapy or experience relapses, eventually progressing to end-stage renal disease.

Chimeric Antigen Receptor T-cell (CAR-T) therapy has shown significant potential in the treatment of autoimmune diseases and may become a promising therapeutic option for patients with refractory or relapsed (r/r) phospholipase A2 receptor-associated membranous nephropathy (PLA2R-associated MN). This study is an investigator-initiated clinical trial (NCT06285279) that enrolled patients with refractory or relapsed PLA2R-associated MN.

Research Conclusion:The study results indicate that BCMA-CD19 dual-target CAR-T cell therapy can induce rapid and durable immunological remission in patients with refractory or relapsed PLA2R-related membranous nephropathy, further leading to clinical remission and improved renal function.

The treatment was generally well tolerated.

This study provides a promising new therapeutic strategy for the application of BCMA-CD19 dual-target CAR-T therapy in refractory or relapsed membranous nephropathy.
