On July 11, the clinicaltrials website showed that Roche initiated a head-to-head Phase III study comparing KRAS G12C inhibitor Divarasib (once daily) with Sotorasib (once daily) or Adagrasib (twice daily) for the treatment of patients with previously treated KRAS G12C-positive advanced or metastatic non-small cell lung cancer (NSCLC).This study is the first to directly compare differentPhase III Clinical Trial of KRAS G12C Inhibitor Efficacy.This study is a multi-center, randomized, open-label, positive drug-controlled clinical trial, planning to enroll 320 patients with advanced or metastatic NSCLC who are KRAS G12C-positive and have been previously treated. The primary endpoint of the study is progression-free survival (PFS).Sotorasib and Adagrasib are currently the only two marketed KRAS G12C inhibitors, launched in May 2021 and December 2022, respectively, with the former from Amgen and the latter from BMS.In addition, there are three other products.KRAS G12C inhibitors have been submitted for marketing approval, respectivelyGlelecis (JACOBIO),Geshuruixi (Zhengda Tianqing/Yifang Biotech) andFuzelase (Innovent Biologics/Gengrui Pharmaceutical). Products from Huajian Future, Merck, Eli Lilly, Novartis, and Roche followed closely, having entered Phase III.
