On July 25, 2024, Roche released its financial report for the first half of the year and updated the latest adjustments to its R&D pipeline, including the termination of four Phase III clinical trials. Among them were two Phase III trials involving the TIGIT antibody Tiragolumab for the treatment of NSCLC: one for first-line treatment combining PD-L1, TIGIT, and chemotherapy in NSCLC, and the other for adjuvant treatment with PD-L1 and TIGIT post-surgery in NSCLC.

At this point, Roche's TIGIT antibody has only five Phase III clinical trials ongoing, two focusing on NSCLC, two on esophageal cancer, and one on liver cancer.

The remaining two Phase III clinical trials around NSCLC are as follows: SKYSCRAPER-01 for first-line treatment of PD-L1+TIGIT in PD-L1 high-expression NSCLC, which announced in 2022 that the study did not meet the primary endpoint of PFS. SKYSCRAPER-03 for first-line treatment of PD-L1+TIGIT in Stage III inoperable NSCLC completed enrollment in Q2 2023.

Clinical Progress on Esophageal Cancer and Head and Neck Cancer

Progress of Phase III Clinical Trials for Liver Cancer and Phase II Clinical Trials for Solid Tumors is as follows.

Summary
TIGIT Antibody Clinical Progress Faces Repeated Setbacks, but Several Companies Developing TIGIT Antibodies Continue Clinical Exploration: Roche, Merck, BeiGene, Gilead, and GlaxoSmithKline Are All in Phase III Clinical Trials.
