On September 11, in order to strengthen academic exchanges on single-cell and spatial omics both in China and internationally, and to promote the in-depth development of fundamental research, clinical research, and translational applications in single-cell and spatial omics, a conference was jointly organized by the Shanghai Genetics Society, the Gene Detection Technology Branch of the China Medicinal Biotechnology Association, the Genetic Diagnosis Branch of the Chinese Genetics Society, and the Translational Medicine Network, among other organizations.MobiDrop, oebiotech, and Illumina Jointly SupportThe "Micro Mastery of All Things - Frontier and Clinical Applications of Single-Cell Spatial Omics Technology Seminar" was successfully held at the Shanghai Cape Hotel.

This seminar brought together numerous industry experts from the field of life sciences research both in China and abroad, as well as entrepreneurs and investors from the industry. Attendees shared the latest research findings, academic discoveries, and application cases, with content covering research achievements, academic findings, and application cases of single-cell spatial omics in areas such as thyroid cancer, breast cancer, prostate cancer, and leukemia, among other hot topics. In-depth discussions were held on how to integrate the two cutting-edge technologies of single-cell and spatial omics to open up broader research fields. The conference not only provided forward-looking insights but also ignited numerous innovative ideas and thoughts, sparking endless creative inspiration.

In the keynote sharing session, Charles Cantor, a tenured professor at Harvard University, an academician of the three U.S. academies, and a foreign academician of the Chinese Academy of Engineering.Academician David A. WeitzLed the opening report. The theme of Academician David's report is“Ultra-high sensitivity diagnostics with droplet microfluidics”, mainly sharing the content of three parts: 1. Creating highly sensitive micro-diagnostic tools using droplet microfluidics technology, with key researchers including Ching Jao and Form Stock; 2. Droplet microfluidics achieves high sensitivity through extremely small reaction volumes, with each droplet containing only picoliter to femtoliter levels of reagents; 3. Compared with traditional microplate technology, droplet microfluidics significantly reduces reagent consumption while enabling higher-throughput reaction processing.

Academician David introduced the application of droplet microfluidics technology in achieving high-sensitivity diagnostic tools. He elaborated on the transformation of replacing traditional reaction plates with droplet reactions, where droplet microfluidics enables a large number of reactions to be conducted in extremely small volumes, significantly improving reagent utilization. Additionally, by constructing microreactors, efficient cell encapsulation and precise control of reactions were achieved. Academician David also provided a detailed introduction to droplet digital PCR technology and its applications in mutation detection, circulating tumor cell counting, and protein sensitivity testing, showcasing the tremendous potential of droplet microfluidics technology in biomedical research.

Chief Physician of the Head and Neck Surgery Department, Fudan University Cancer HospitalWang YulongProfessor"Microenvironmental Changes and Translational Research in Thyroid Cancer Initiation and Progression"Prof. Wang shared on the topic. He mentioned the high incidence and severe prognosis challenges of thyroid cancer. He conducted an in-depth discussion on translational research regarding microenvironmental changes in the initiation and progression of papillary thyroid carcinoma (PTC), particularly using single-cell technology to analyze PTC cells, offering new perspectives for clinically confirming evolutionary pathways and predicting treatment efficacy. Meanwhile, he revealed the phenomenon where TERT promoter and BRAF V600E co-mutations exacerbate the malignancy of thyroid cancer. Through single-cell sequencing, the team discovered that although undifferentiated carcinoma has a high mortality rate, it is sensitive to immunotherapy. The combined application of immunotherapy and targeted therapy significantly extends the survival period of patients with undifferentiated carcinoma. Additionally, the research team found that the BPC cell line can be used for experiments in mice with normal immune function, possessing unique characteristics of the thyroid organ, ushering in the immunological era of thyroid cancer research. This study addresses the most fundamental needs in thyroid cancer research. Although similar mouse model studies exist internationally, there has been no successful cultivation of reproducible cell lines. This research resolves the "bottleneck" issue in thyroid cancer research in China.

Xinhua Hospital Affiliated to Shanghai Jiao Tong University School of MedicineSong JiaAssociate Researcher Shared a Presentation Titled"Single-Cell Spatiotemporal Multi-Omics Sequencing Analysis of the Tumor Microenvironment"Professor Song mentioned in his report that their team has developed the Well-TEMP-seq technology by utilizing the chemical tool of RNA metabolic labeling, which, combined with single-cell transcriptome sequencing technology, enables efficient analysis of gene expression dynamics in thousands of single cells.

Subsequently, the head of Illumina's Americas Research Applications MarketRobert YamullaDr., the topic of his report is“One flow cell, multiple omes: proven supported methods for CRISPR, single cell, circulating RNA and more”He stated that Illumina is integrating multi-omics workflows and scientific discovery ecosystem solutions. The NovaSeq™ X Series, Illumina's flagship sequencer, represents a significant leap forward in sequencing innovation. Its advantages include: more samples at a lower cost; flexible workflows; reduced packaging waste and lower consumable costs; automation and ease of use; and higher accuracy. With NovaSeq™ X, the scientific community can achieve single-cell and spatial omics sequencing at a more affordable price. He elaborated on the excellent data performance of single-cell CRISPR/Cas9 screening on NovaSeq X and shared several international research and innovation collaboration cases based on NovaSeq™ X.

Vice President of Fudan University Shanghai Cancer CenterJiang YizhouThe research fellow's report topic is"Molecular Subtyping and Precision Treatment of Triple-Negative Breast Cancer"Professor Jiang shared that their team, based on an in-depth metabolomic study of 330 triple-negative breast cancer samples and their paired tissues, successfully mapped the largest global metabolite atlas of triple-negative breast cancer to date. The study revealed metabolic characteristics such as significantly upregulated phospholipids and sphingolipids, along with the accumulation of glycosylated metabolites, closely linked to tumor genomic alterations. This research further categorized triple-negative breast cancer into three subtypes: sphingolipid-enriched, oxidative and glycosylated metabolite-enriched, and low metabolic disorder, optimizing the "Fudan Classification." Professor Jiang particularly noted that the further classification based on metabolic features not only improved the predictive accuracy for the prognosis of patients with the basal-like immune-suppressed subtype but also uncovered potential metabolic therapeutic targets. Additionally, it was found that the luminal androgen receptor subtype showed a high overlap with the sphingolipid-enriched subtype and exhibited ceramide lipid accumulation, potentially opening new avenues for the precise treatment of triple-negative breast cancer.

The next speaker is the General Manager of Oebiotech Biomass Spectrometry (Luming Biotech).Shu LieboDoctor, with"New Approaches in Cohort Studies: From Classic Omics to Single-Cell and Spatial Omics"Dr. Shu shared the Level One cohort metabolomics customized system, and the team has also developed a distinctive functional metabolomics database. The team is advancing the work of cohort screening and translational applications, and Dr. Shu shared representative collaboration cases. Meanwhile, the team has developed Deep Blood Full-Spectrum Proteomics detection and analysis. Based on this new method, the team has increased the protein extraction results to 4000-6000. In addition, he introduced the three major platforms of single-cell omics, single-cell omics analysis, the spatial metabolomics system, the exclusive qualitative database for spatial metabolomics, the application expansion of spatial metabolomics, spatial transcriptomics, research achievements in spatiotemporal multi-omics aiding tumor molecular subtyping, and new models of spatiotemporal multi-omics applications.

The seventh speaker is from Ruijin Hospital, Shanghai Jiao Tong University School of Medicine.Yin TongResearcher, Professor Yin Focuses"Application of Spatial Metabolomics in the Study of Metabolic Reprogramming in Acute Myeloid Leukemia"The unique advantages and platform differences of spatial metabolomics technology were elaborated in detail, and the technical principles of AFADESI-MSI were thoroughly analyzed. Through an AML case study, she demonstrated the practical value of this technology in clinical settings, revealing the complex mechanisms of tumor cell metabolic abnormalities independent of genetic mutations. In addition, Professor Yin introduced the establishment background and project positioning of the National Center for Translational Medicine at Ruijin Hospital. The construction, operational effectiveness, and research output achievements of the National Major Scientific and Technological Infrastructure for Translational Medicine (Shanghai) and the Ruijin Base Mass Spectrometry Platform were also presented. She shared collaboration cases between the platform and the respiratory department (analysis of immune characteristics of Omicron-infected individuals vaccinated with inactivated virus), pancreatic cancer (proteomics-driven precision treatment of PDAC), as well as LDT projects (quantitative detection of plasma venetoclax concentration).

Special mention was made of the successful application of the MobiNova®-M1 system in microbial genomics research, such as revealing the relationship between cancer and strain genes and the evolution of pathogenic microbial resistance, as well as the challenges and breakthroughs of MobiMicrobe scRNA-seq in studying the impact on microbial community evolution.

The last guest to share is from Fudan University Shanghai Cancer Center.Wang JianhuaResearcher. He conducted"Application of Single-cell and Spatial Multi-omics Technologies in Prostate Cancer and Coal Worker's Pneumoconiosis"Academic Sharing. He stated that prostate cancer ranks second in incidence among male tumors in China and has been continuously rising in recent years, with 120,000 new cases in 2020. It is expected to further increase with the aggravation of aging. Its pathogenesis is complex, involving factors such as race, individual malignancy, androgen dependence, and cellular heterogeneity. To further explore the prostate cancer microenvironment, the research team used single-cell sequencing technology to divide the prostate tumor microenvironment into nine cell subpopulations and combined the advantages of spatial transcriptomics to reveal intercellular spatial relationships. The study found that specific cell subpopulations are closely related to the progression of prostate cancer, and prostate cancer-associated CAFs have relatively high tissue specificity. Additionally, about one-third to forty percent of prostate cancer patients exhibit abundant immune cell infiltration, particularly T cells. In recent studies, the team revealed the heterogeneity of coal dust lung progression in mice and a vitamin D replacement approach through single-cell sequencing. Vitamin D3 reverses immune tolerance and enhances the cytotoxicity of effector T cells in coal dust lung.
