Today, AstraZeneca and Daiichi Sankyo announced that their jointly developed重磅 antibody-drug conjugate (ADC) Enhertu (trastuzumab deruxtecan) achieved positive results in the phase 3b/4 clinical trial DESTINY-Breast12.Enhertu Demonstrates Significant Overall and Intracranial Clinical Activity in Patients with HER2-Positive Metastatic Breast Cancer and Brain MetastasesThis result was released at the European Society for Medical Oncology (ESMO) Annual Meeting and simultaneously published inNature MedicineUp.

The test results showed,In patients with brain metastases at baseline, the 12-month progression-free survival rate assessed by independent central review was 61.6%. Additionally, the 12-month intracranial progression-free survival rate for patients with brain metastases was 58.9%.In patients with stable and active brain metastases, the results were consistent. The 12-month progression-free survival rate for patients with stable brain metastases was 62.9%, and the intracranial progression-free survival rate was 57.8%; for patients with active brain metastases, the 12-month progression-free survival rate was 59.6%, and the intracranial progression-free survival rate was 60.1%.
In patients without brain metastases at baseline, the objective response rate (ORR) assessed by independent central review was 62.7%, including 23 complete responses (CR) and 128 partial responses (PR).See the table below for specific values.

Enhertu is an ADC therapy jointly developed by AstraZeneca and Daiichi Sankyo. It is designed using Daiichi Sankyo's proprietary DXd ADC technology platform and consists of a humanized monoclonal antibody targeting HER2 connected to a topoisomerase 1 inhibitor payload via a cleavable tetrapeptide linker.Enhertu was first approved by the U.S. FDA in 2019 for the treatment of patients with unresectable or metastatic HER2-positive breast cancer.
