On September 20, 2024, Eli Lilly registered a Phase I clinical trial for LY3537031 in weight loss treatment on Clinicaltrials.gov.

According to previously disclosed information from Eli Lilly, LY3537031 is a GLP-1R/GIPR dual-target agonist that has undergone a Phase I clinical trial for glycemic control. This Phase I clinical trial targets weight loss indications, planning to enroll 230 subjects, with an expected completion date of July 2025.

LY3537031 is still administered via subcutaneous injection. Eli Lilly has advanced three GLP-1/GIP dual-target agonists into clinical stages, including tirzepatide and two others, LY3537031 and LY3493269 (subcutaneous or oral formulations), which are being developed for type 2 diabetes indications. However, no clinical data has been disclosed for these two, and after completing phase I trials, no further development was observed. The restart of clinical trials for LY3537031, now pivoting to a weight-loss indication, signifies that the second GLP-1/GIP drug has re-entered active development.

Recently, it can be seen that Eli Lilly and Company is fully advancing the clinical development of multiple dual-target/multi-target GLP-1 pipeline drugs. In addition to the already marketed tirzepatide (the high-dose version is in phase II clinical trials for people with severe obesity BMI≥35), the three-target Retatrutide and Masdupeptide have also entered phase II clinical trials. A few days ago, data from the phase I clinical trial of the 16mg weight loss dosage was disclosed, and the second GLP-1/GIP drug has also restarted clinical trials for weight loss treatment.
