Recently, Lepu Biopharma Co., Ltd. ("LEPU BIOPHARMA") and DP Technology announced the achievement of a significant milestone in their ADC drug collaboration. This partnership has deeply integrated DP Technology’s ADC Linker-Payload design platform with LEPU BIOPHARMA’s ADC technology development platform, leveraging each other's strengths. Through multiple rounds of Linker iterative design and validation, the collaboration has optimized the overall drugability of ADCs within a relatively short period. Recently, this project achieved a key milestone, further validating the pathway of accelerating ADC innovative drug R&D through computational design. Moving forward, both parties will continue to deepen their R&D cooperation in this field, jointly committing to advancing and optimizing the process of innovative drug development.

The ADC Linker-Payload Design Platform, meticulously developed by DP Technology, is built upon the "RiDYMO® Difficult-to-Drug Target R&D Platform" and further integrates "an AI + First Principles-based computational approach for predicting linker cleavage sites." This ensures not only the precise release of the designed payload but also achieves excellent cell-killing effects and a "bystander" effect. Meanwhile, the designed antibody-drug conjugates (ADCs) exhibit superior hydrophilicity and plasma stability while maintaining high drug efficacy. Supported by LEPU BIOPHARMA’s mature and comprehensive ADC development system, the new ADC platform has demonstrated significant superiority in CDX efficacy experiments targeting multiple antigens. This project has been effectively validated in animal models, and candidate molecules are currently being developed for clinical-stage advancement.

Currently, in response to various needs from industry partners in the ADC field, the DP Technology platform has successfully empowered multiple scenarios and projects. Examples include using AI + first-principles calculations to predict Linker cleavage sites and enhance attachment stability; leveraging pre-trained models combined with fine-tuning strategies, along with expert experience, to predict and modify Payload efflux, bystander killing, and other physicochemical properties; and continuously exploring and collaborating on the evaluation of overall ADC properties such as aggregation effects.
