On October 10, 2024, local time, Pfizer announced significant news in the field of prostate cancer treatment. The combination regimen of its poly ADP-ribose polymerase (PARP) inhibitor Talzenna and androgen receptor pathway inhibitor (ARPI) Xtandi achieved remarkable results in the study for the treatment of metastatic castration-resistant prostate cancer (mCRPC).

According to the topline analysis of the Phase 3 TALAPRO-2 study, the combination regimen improved overall survival (OS) in patients. The results showed that, compared with Xtandi alone, the final overall survival (OS) improvement was statistically significant and clinically meaningful for all participants as well as for mCRPC patients with homologous recombination repair (HRR) gene mutations. At the time of the final analysis, radiographic progression-free survival also maintained a clinically meaningful improvement. This outcome makes Talzenna combined with Xtandi the first and only PARP inhibitor combination used with ARPI that significantly improves survival rates in mCRPC patients, regardless of mutation status.
Previously, in June 2023, the FDA approved Talzenna in combination with Xtandi for the treatment of adult patients with mCRPC and HRR gene mutations, and the EU also approved it in January 2024 for adult patients with mCRPC without clinical indications. Currently, this combination therapy has been approved in more than 35 countries/regions worldwide for the treatment of mCRPC patients.
In terms of safety, the adverse events in the TALAPRO-2 study were consistent with the known toxicity profiles of the individual drugs. Detailed results will be presented at upcoming medical conferences, and Pfizer will share the data with global health authorities to update and expand the approved labeling for Talzenna.
Prostate cancer, as the second most common cancer among men globally, has its most advanced and aggressive stage known as mCRPC. The progress in Pfizer's drug research not only brings new hope to patients but also draws significant attention from the industry regarding the company’s management dynamics. With further promotion and application of subsequent research results, there is potential to open a new chapter in the field of prostate cancer treatment.
